| ...with DNA samples should always have the following:? During electrophoresis, gels that are run with DNA samples should always have the following: (1) an unamplified control lane (i.e., a reaction tube without any DNA sample); AND (2) a lane with a DNA ladder. a. What would be present in the unamplified control lane? What would you expect to see in that lane after the gel is run? Explain. b. What is a DNA ladder? What would you expect to see in that lane after the gel is run? Explain. c. Why would these two extra lanes be helpful when analyzing your gel? |

| (1) an unamplified control lane is a pure RNA without RT made. It serves to visualize genomic DNA contamination. If no contamination, nothing should be seen. (2) a DNA ladder is a ladder made of 100 base pairs so you can better visualize your PCR products, according to the size. After the run, they should look like a ladder, with a band every 100 bp.
