| Hi, In my thesis I will isolate the coding sequence of a particular gene from a plant, I determined the conserved regions of the gene from other organisms and I desing primers. I isolated mRNAs and made cDNAs, after PCR, 600 bp long fragment amplified. I made it sequenced, and the fragment is %95 similar to the genes that have been isolated from other organisms. So, here I have two choices to obtain the full lenght cDNA, I can construct cDNA library and screen the library for the clone I am interested or I can do RACE PCR. Which one do you think would be easier? And if you have experience with these techniques could you tell me the advantages/disadvantages? Thank you... |
