| HaeIII recognizes the following sequence: 5'...GG/CC...3' 3'...CC/GG...5' If your DNA is short and linear, the number of band will be one plus the number of times the above sequence appears (assuming your enzymatic cleaveage reaction worked properly). If your DNA is circular, you will get the same number of bands as you have instances of the above sequence. (usually the band due to circular DNA shows up elsewhere) Also, look at your bp ladder control lane to get an idea of size of the fragments, and try to correlate that with the expect cuts based on your DNA sequence. |
