| Hello all, I'm working with whole blood and isolating plasma/buffy coat from EDTA treated tubes. I'm centrifuging the samples to isolate the layers based on density and pulling the plasma and buffy coat respectively. From here I will be pelleting the buffy coat and storing the pellets and plasma in -80 for later DNA/RNA extractions. I am using the Fujifilm autogen to carry out extractions. I am curious how many extractions I will be able to carry out using a single buffy coat pellet. The required cell count for a single sample prep is 3 x 10^6 suspended in 200 micro-liters pbs. Any help or suggetions would be greatly appreciated. thanks and cheers, Matthew |
