| I have been looking for a binary vector and have only found pGreen and pCLEAN. After looking into both of these vectors, I decided pCLEAN would be the best for me. So right now I am trying to figure out which vectors I want and came across some interesting information. In a article evaluating pCLEAN vectors I saw that trans-genes located out side the T-DNA region (LB and RB borders) were being integrated into plant cell DNA. ![]() ![]() -pulled from [Only registered users see links. ] Legend- ![]() -pulled from [Only registered users see links. ] How does this occur, and does it occur naturally with a regular Ti-plasmid? While you are viewing this, what vectors would you recommend I use? I am trying to engineer a plant expressing GFP for strictly visual representation of Agrobacterium-mediated gene transfer. I know what genes I want (GFP, HPT and Kan-R/Tet-R), but for the pCLEAN-S plasmids I am confused on why some of them contain T-DNA. What benefit is it to have two T-DNAs?
