| Hey Everyone, I have a pSMPUW vector that is low copy number. ORI seems to be ColE1 derivative with point mutation in ROM/ROP. I have compared the ORI to the High Copy Number pUC ORI in the PLVX vector and the only difference between the two is a three base pair deletion of TGG in PLVX. pSMPUW- GTAGCGGTGGTTTTT PLVX- GTAGCGGTTTTT Does anyone know if I do mutagenesis to delete the TGG nucleotides if it will create the same high copy number of the PLVX plasmid?
