| hai..im new here..im interestd in studying about gene expression of exon. howevr, im not sure which source of DNA i should use..cDNA or genomic DNA. which is the best and easiest source for good results. is it possible to amplify sequence of 4000bp? and what are the materials needed for maximum products? |

| Since you're only interested in the coding bits then cDNA would be best unless you're going to collect mRNA. Under 10 kb can all be amplified pretty much the same. You do want your extension time to be about 4 minutes per cycle (about 1kb/minute). Make sure your primers have a melting temperature within a couple of degrees of each other and start with your annealing temp about 4 degrees below that melting temperature. |
