Post Proteinase K



Hello! I am studing fetal DNA and I usually use a Qiagen kit (wiht silicagel membranes) for the extraction, but Iīve run out of Proteinase K and the lysis buffer. So I thought I could make it myself, but I canīt find one thing. I donīt know how to relate the activity of the protease with the concentration (mg/mL). Does anyone can help me? and if you have a home made protocol for extraction of DNA using silica gel membranes it would be very helpful. Thank you!!!!

Hi Carobiotech, usually buffer don't run out so fast compare to column (in your case silica gel membrane).

check this out

it give you the activity of the proteinase K used... for the lysis buffer ATL... if you look hard enough, you should be able to find that ATL is a physiological buffer (well, I could be wrong... its online somewhere)
