| I am going to extract Phytophthora fungi DNA from wood samples and medium agar colonies. I have some experience with plant tissues, like leaves and needles with Macherey Nagel NucleoSpinŽ Plant kit. It worked really good. With fungal DNA I have no practice at all. I have contacted the manufacturer and received a vague explanation that they did not tested Phytophthora extraction, but they think that it is possible if the material is well-homogenized. I will use liquid nitrogen for homogenization. For extraction of other fungi they propose a modified protocol by adding sea sand for better homogenization and a chloroform. Here I have a dilemma: should I use this modified protocol with sand and chloroform only for extraction from mediums or also for wood? Does this modified protocol gives only better yields of DNA in comparison to the standard one or no DNA will be extracted without chloroform at all? Any suggestions would be very valuable.
