Default DNA Extraction from Yeast



Hi All,

I'm hoping someone might be able to help me. I am trying to isolate DNA from fresh yeast but keep getting products with really low DNA concentrations (around 25ng/ul). I'm using the NucleoSpin Plant II kit from Macherey-Nagel, which I have used before with other fungi without any trouble.

I'm stumped, does anyone have a suggestion?

I have tried to obtain DNA from methanogenic bacteria, use the BIOMASS kit, without success, any suggestions?

Hi Jspall,

Did you used the basic nucleospin plant kit protocol or the second one adapted for fungi? It seems to be a problem with good homogenization of your samples, isn't? How do you performed a homogenization?

when extract DNA from yeast, the concentration commonly is low,It's not your problem
