| Hello people, It's been a while, but it's nice to see you guys are all alive and kicking. My question: Is it ok to cut out the band of interest in the agarose gel and leave it overnight at 4oC to extract tomorrow? Thanks |

| Some kits recommend to freeze it at -20º C, so I think there would be no problem leaving it at 4ºC |

| DNA in an agrose plug (essentially what a gel slice is) is extremely prone to nuclease attack. Especially if you just happened to use any old gel box and reagents (just normal everyday contamination). So leave the plug in 25 mM Tris pH 8.0; 50 mM EDTA. And it'll be stable at 4 C for a long long time. BUT the smaller your DNAs are the more likely they are to diffuse out into the buffer over time. But really really if its just O/N before extraction you should be fine. If the DNA is precious and you can't risk any loss just process the buffer (use just enough to cover the plug while storing to keep volumes low) too during extraction. |
