| I am digesting lambda DNA using a restriction enzyme, which will give three fragments. I want to separate them for subsequent use. I am trying to extract them from agrose gel and then purify using gel extraction kits. However, the yield is really poor. Are there any other easy and efficient ways to separate them? Or, please recommend me some excellent gel extraction kits you knows. Thank you very much. |

| Lamba DNA has sticky ends so needs to be heated before running on a gel. A long time since I did this, but from memory 95 degrees C for 5 mins should be OK? For the gel extraction, if you want to contact me I can give you more advice without making a really long post. In general the Machereny-Negel Nucleospin II lit is good, but I have optimised the protocol. |
