Unhappy Problem in plasmid extraction



Hi everyone, I'm Alston. I'm doing plasmid extraction using alkaline lysis method. But the extracted plasmid can't dissolve in TE buffer (pH 8.0) although I pre-heat the buffer. Or may due to other factors during the whole procedure?
Anyone can help? thanks

If you are having the problem with TE buffer then you can dissolve it in Sterile MQ water. It will with work same efficiency. I never dissolve in TE buffer.

Hi Alston,

I can't help you with your problem, but I want to do plasmid extraction myself, but have never done it before. So I don't know how it works and was wondering if you could give me some advice in how to get a bacterial culture and how to extract their plasmid DNA? Also, is there a way to know the DNA sequence, because I want to use the plasmid DNA in PCR, but need to design primers for it.

I hope you can help me.

From limited experience i doubt its the buffer. Do you mean dissolve or elute from a column?
