Can anyone give me 2 ways in which you can enhance the resolving power of the microscope?

And also what objective focuses closest to the slide?

Can anyone give me 2 ways in which you can enhance the resolving power of the microscope?
1. adjust the iris diaphragm to smaller, but this may cause insufficiency of brightness expecially in fluorescent image.

2. Using a higher power objective. but this will cause a decrease in field of view.

And also what objective focuses closest to the slide?
Do you mean liquid immersion objective lens (such as water or oil immersion lens)? I am not quite understand your question!

what is a refractive index
The refractive index (or index of refraction) of a medium is a measure for how much the speed of light (or other waves such as sound waves) is reduced inside the medium. (information from Wikipedia)
