What is the best strategy to purify an unknown protein using HPLC?

uv detection at 280 nm, use a c18 column, use a fraction collector and collect by peak fractionation..

sorry...i have another opinion~~
instead of using 280nm, can cawhy dont we use PDA to screen a range of different wavelength then for fractions colelction? we wont miss any that might not be/ less active under a that particular wavelength ?

correct me if i making wrong statement~~thanks
