Default Spherisurb SAX column problem

Hello,

I am utilising the following column and I'm experiencing problems with it:

Spherisurb SAX 5 um 250x4,6.

I presume the column is okay, and the washing method is not.

Can anyone help me out by telling me what washing method to use?

Hi,
What kind of "molecules" you want to strip?Cations of metal, organic hydrophobic and hydrophilic molecules, organic cations?

Here is the Phenomenex recommended column washing procedure for IEX columns:

Rinse w/ 10 column volumes (CV) each of:
500mM phosphate buffer pH7
10% acetic acid (aq)
5 CV of water
10 CV of phosphate buffer pH7
5 CV H2O
10 CV MeOH
10 CV H2O
For protein removal repeat above with exception:
replace 10 CV MeOH wash with 10 Cv 5M Urea or guanidine thiocyanate

In our lab we use a stripped down procedure which seems to work OK when analyzing simple mixtures -
10 CV phosphate buffer pH7
10 CV H2O
10 CV MeOH (for storage)
