Intact Mitochondria isolation

I found a protocol which is adapted from Attardi and Ching (1979) and Rice and Lindsay (1997), is designed for cultured cells (2.10^8) grown as a monolayer. It's really close to your protocol. I hope it will be enough pure for my EM experiement

Ok i have succeed to obtain mitochondria, apparently my problem was the concentration of cells, I increase by 3 the number and that's work with differential centrifugation. Thank you very much for your help. I have a last question do you know what kind of compound can I use to replace the sugar in the buffer ( sucrose and manitol) and keep the same osmolarity.

You may be able to do it with a protein like BSA; however, you'll need a osmometer to accurately measure the osmolarity.
