Default medium for growing HEK cells in low pH (~6)



I need to grow my HEK cells in an culture medium with a relatively low pH (6-6.5). They are currently grown in RPMI-Medium (+ 10% FCS + several other supplements). I tried to adjust the pH to about 6.5, but after growing the cells for 2 days, the pH rises to more than 7.0. I think the medium is HEPES-buffered, what I think could be the problem. But I don't know what other medium to use. Do you have any suggestions? (Or is it possible to regulate pH by CO2? (currently 5%))

I think you are growing HEK cells in shake flask probably, i grew CHO cells,and normally its pH fall from 6.9 to 6.5-6.6, so i do not know why your pH rising. but one can be congsidered: when you sampled and measure the sample's pH,the value you got will be higher than the real pH value, just because the disolved CO2 in the sample run to the atmosphere.
