Cell Mortality: Please Help me
I haves a big problem for my cell culture recently .
I'm culturing the adherent mouse macrophage cell line Raw264.7 for 2 years and it goe's well until now.
The problems i'm facing now is poor cell morphology and viability They are adherent as they are supposed to be, but we saw a lot of mortality, a lot of vacuoles in cells and many lysis areas with large size cells (that seems apoptototic). I haven't any bacterial or fungal contamination that can be visually observed under the microscope.
I have used new: RPMI(SIGMA), FBS(Hyclone), Tissu culture flask and i didin't seen any improvement.
I wonder whether there is any perceptible signs for mycoplasma contamination or another type of contamination that I can use to judge my case?
It drive me crazy !!!
