Default Mycoplasma contamination?



Dear all,

I got a big problem for my cell culture recently, the S1 (HMT-3552) refuses to behave themselves in 3D culture (in Matrigel). Normally, they stop proliferating (exit from cell cycle) after day7 when I remove the EGF, however, they start to continue growing after day 7 recently. And some lab members doubt that we may get mycoplasma contamination, however, I did not see any changes for the medium (it is not turbid but as clear and transparent as normal).

I wonder whether there is any perceptible signs for mycoplasma contamination that I can use to judge my case?

Probably, there are no straightforward ways to detect a mycoplasma contamination.

However, for this purpose there is the Hoechst staining solution (Sigma, cat# H6024). Unlike other methods, there's no need to perform PCR, even though you need a flourescence microscopy.

Have a shot at Lonzas mycoalert for a really easy mycoplasma test, it is quite expensive, but as its an enzyme based assay its real easy, I've never seen turbidity when a cell line is contaminated with mycoplasma more a change in morphology or decreased growth rate
