Hello serum is used as a nutrient for the cells ie Fetal bovine serum

Passage is splitting/or cell trypsinization and then subculture.

1. for cell , nutrition and growth factor are necessary to grow
2. we culture cells on dish or plate, when cells grows enough to cover the whole dish or plate, “contact inhibition” happen and cells may stop to grow, in this case we need to divide the cells from the original dish or plate to new dish or plate to keep the cells growing. this process we call it "passage"

if my explanation have any mistake, please revise it. FYI.
