Arrow Cell Passage Number for Cell Culture

Dear fellows,
what is the maximum cell passage number during subculturing? I have heard this is 30, however what is the optimal cell passage number?

Does this also depend on the cell line?

Every cell line is different. Established cell lines generally can be passaged more than primary cells. I have passaged J774 Mouse Macrophages over 200 times without problems. The most important thing to check is that the receptor/cytokines/enzyme/protein that you are interested in is STABLE over that period of time.
We have used endothelial primaries and found that certain prostaglandins change almost instantly, within 1 or 2 passages. Make sure to have banked early passage cells so if there are changes, you have cells to re initiate.
