Question Cell Adhesion Problem After Transfection

Hello guys, I have another problem....


This time its cell detachement after transfection. Can someone help?:
  1. I am seeding HEK293 (260,000 cells/well in the dish) in 24-well plates.
  2. I transfect 24h post seeding. I am transfecting using lipofectamine.
  3. Cells grow fine. I wash them with 1mL HBSS (Gibco) and incubate them for 10 minutes before transfection. Cells looks fine, but seem a bit stressed after the incubation.
  4. I add my transfection mixture, and right after transfection the cells start to detach from the bottom of the plates.
I now have changed the media to Optimem, got new and better plates, and nothing helps!

Hello there

it may be you are transfecting with too much DNA / transfection agent or your cells may not be healthy.

Have you tried this again or optimized your transfection?
