Default Marsupial chromosomes

Hi everyone,
I'm doing some cultures of kangaroo lymphocytes (72h/37ºC) and after arresting them with 0.5 microgram/ml colchicine (1h), shocikng them with o.o56 KCl, and fixing them in methanol:acetic acid I don't see a single metaphase. Not even bad looking metaphases, as if the colchicine exposure were too long.
Any clue?
