We weaned cell cultures off serum over a 5-7 day period. Our first time doing this. It worked very well, going from 100% usual serum to 75% to 50% to 25% to 0%. We even asked ourselves if going from normal serum media to serum free media in one step could have been possible, but we didn't bother to investigate. The cells seemed impervious to the media changes, no morphological, dying off, no slower replication absolutely no difference. They lived and divided for a month until we stopped culturing and collected the cells.
