If any one could answer my question i will be happy, What could be the reason for getting efficiency around 60% in my real time pcr, i am using SYBR green I dye. In the same run some sample showing 100% efficiency while most of other samples showing only 60-80% efficiency.

Dear all,
I have problem with detecting P65 through western blot. when i run the sample i can find the band for first time but when i reapeat it with the same sample i could not detect the band.

I am sure about storing the samples and using the Protease inhibitors. What else could be the problem.
