Pcr Problems

Posted 11-13-2008 at 04:58 PM by andrear
Hi - I need help figuring out what is wrong with what I'm doing for PCR...
I have two primers that were designed but they are rich in cs and gs.
Anyway, aside from that problem the gel shows that the bands are too small for what they should be
how do I get the bands to be bigger?

    Hi andrear,
Good morning, I am not an expert in pcr but still i am doing it for quite some time so i am assisting you with what i have learn. The prob that you were facing with the pcr is you are not getting the correct pcr products, for me it seems like the extension time of your pcr run has to be rechecked because the bigger the product size you might have to extend you extension time, i suggest you to play around with extending the extension time usually people will use 15second but that was the default it will vary according to the length of the strand that you are amplifying. I hope that i answered your doubt.
