Default Ligation - too low concentration of DNA



Hi.

After purify DNA from gel, I got very little of DNA concentration about 1 to 2 ng/ul. Can I do cloning with this amount of DNA? Can anyone give me any tips to increase the DNA concentration? Thank you in advance.

Hi Nurul
if you have a large volume of DNA you can run it through a column or filter and elute only with a small amount of volume. Eg run through 200 ul but elute with only 15 ul, Then you can use the sufficient ng for cloning

By the way if you clone well with DNA Ligase and transform with highly competent good cell number you should get clones even with 2ng likely
