| Hello, I am using transgenic mouse where my gene of interest is floxed. When I cross this mouse line with Tamoxifen inducible Cre line, CAGGCre-ERTM, I get mice with with heterozygous Cre and floxed gene of interest. The experimental plan was to isolate myoblasts from these mice and induce Cre in cell culture by addition of Tamoxifen. To my surprise, the cells isolated from these mice die with in few days, where as the cells from WT or Flox/flox mice do not. Could anyone tell me what could be the problem here and also give me some suggestions on how to bypass this problem. Thank you... |
