| Procedure to destain PVDF (polyvinylidene difluoride) membrane stained with Ponceau S : After proteins are transferred onto membrane, immerse the membrane in Ponceau S staining solution and stain for 5 minutes. After proteins have been visualized, rinse the membrane with distilled water and rapidly immerse in an aqueous solution of 0.1 M NaOH. Protein bands will start to disappear after 10-30 seconds. Rinse membrane with running distilled water for 2-3 minutes. Continue with other procedure being followed (e.g. immunological detection). |
