Default give the information about primer concentrations



i have primer of 45 nmoles of conce. how much i will use for pcr. and how i will dilute. plz

Hi Muhammad, what is the final conc of primer conc you are expecting or plan to use? If you know just use the equation M1V1=M2V2.

at the moment you know M1 which is 45nM.
V1 = initial volume
M2 = final conc
M2 = final volume

10x or 10nM?

Let us look if it is 10nM (you can calculate yourself if other conc require by looking at example below)

M1V1=M2V2
M1 = 45nM (initial stock conc)
V1 = X, denote volume require of the stock
M2 = 10nM conc to archieve
V2 = 25uL, final volume to archieve

put it all in you get

(45)*X=(10)*25
rearrangement
X=((10)*25)/(45)
X=5.5555, or 5.56uL

Thus in a 25uL reaction you require to pipette stock primer volume of 5.56uL.

**Important, when using M1V1=M2V2 make sure you SI unit is the same for volume and conc.
