Question How do I determine size of calf thymus DNA cut with Hind III?



For this question I was asked to draw up a calibration curve and determine the average size of the fragments of calf thymus DNA after cutting with Hind III and Hae III...

Is it possible for anyone to explain to me who I go about determining the average size of these fragments using my curve? I am not sure how to go about doing this.

I usually take my DNA and throw it into Webcutter.

The size and type of your DNA may cause issues as it is huge. Is there anything in particular you are interested in ie gene? Just add that to webcutter

Not particularly I find it interesting but wont be doing a thesis on it or anything :-) It's just a simple investigation for university. I am slightly confused about how to figure out the cutting frequencies of these two restriction enzymes (Hind III, Hae III) in calf thymus DNA though. I used agarose gel electrophoresis and now I am trying to analyze the gel image results. It's difficult as I can't differentiate between the bands... it's just a smear?

Hi there would be hundreds if not thousands of cuts.
Calf Thymus genomic DNA would be about the same as calf genomic DNA so that may even be millions of cuts.

You can estimate this based on finite math/stats

Find the likely hood of the match of 6 bases in a row

1/4 (A or G or C or T) - 6 times

or 4X4X4X4X4X4X4 or 4^6 but there are billions of bases in sets of 6, chances are there are many spots. I can post the exact answer later.
