Default Positive Colony Screening



Currently, I have done a transformation and there are a lot of colonies growing on my culture plates. Now, I need to find which colonies contain my desired insert. As I know, I can pick up those colony bu using toothpick then how I can make it by using PCR screening for the next? The primers are needed? any method for colony screening?

How to make my desired genes which still shows clearer bands after a series of purification? It usually becomes fairer after I extract the gel and seems nothing dna or maybe too little genes are established at the next gel running.

Reecently, I have done the double digestion test on my plasmid. But it may got wrong because I cant see any bands shown on gel electrophoresis after 5 hrs or overnight incubation on 37C. Then I tried to test one enzyme by one enzyme by using singke digestion, they are worked & fairer bands are shown. why this can happen?
