 In column chromatogaphy, if i would like to increased a colume volume of... ...200ml how could i do so? Ok mark the 200ml volume at the empty column. Then i pour in the silica gel and followed by the solvent to dissolve the gel. If i see the gel touch the 200ml mark immediately after mixing with the solvent, does this confirm the gel volume has already been achieved?Or do i need to wait for the gel to settle?Does the gel volume differ when it settles or is it the same before it settle?But after i wait for it to settle, if it doesnt reach the 200ml mark lets say it only reaches 150ml, what can i do, i can add anymore gel rite after it settles?How long does it take for the gel to settle?  |
