| amplicons(gene of interest) are ligated into plasmid vector..thus i assume the plasmid's ability to produce b-galactosidase is disrupted..but this plasmid-amplicon thing is transformed into e.coli cell and grown on LB agar.. so whether the colony is blue/or white is because the plasmid lac z becomes disrupted OR the e.coli lac z becomes disrupted? my guess is the the e.coli used is treated and cannot produce b-galactosidase in the first place |
