Hello, new here and thought I'd make my first post with something that has been troubling me quite a lot at uni. It's to do with bacteriophage suspensions, and colonies / plaques formed etc. I just can't seem to get my head round it, and would greatly appreciate any help / websites that would help me understand better.
Here is an example of a question recently set. I'm not looking for answers or to be spoonfed, more just pointers about how I'd go around solving such a question and help around the general area.
100 microlitres of a thousand-fold diluted bacteriophage suspension was mixed with 0.2ml bacterial suspension and plated on a petri dish. 258 plaques were formed. |
A) The titre of the original phage suspension was 8.6 million pfu/ml - True or False
B) The titre of the original phage suspension was 2.58 million pfu/ml - True or False
C) The titre of the original phage suspension was 25.8 million pfu/ml - True or False
D) To obtain 5000 plaques per plate you should mix 0.2ml cells with 193 microlitres of a hundred-fold dilution of phage - True or False
E) To obtain 5000 plaques per plate you should mix 0.2ml cells with 57.9 microlitres of a thousand-fold dilution of phage - True or False
Now I'm pretty sure all these questions revolve around one simple calculation, which if provided is great but it's still information on the general area that I'd like as well. Any help would be appreciated, cheers.
