my question is how to isolate the proteins by ultracentrifuge ? thanks

Ultracentrifugation (UC) requires a very pure sample. I haven't heard of it being an isolation technique by itself.

UC measurements are performed by high level optics during spinning

One you stop the sample, equilibrium would mix your sample therefore it would be hard to extract only your desired protein.
