Default Glycerol Stock



I have worked on plasmid DNA isolation and cell retransformation and i was ask to prepare glycerol stock for recombinant strain and how to ensure it is functional of gene expression.

For preparation of glycerol stock: I chose -70oC with 15% glycerol.

However, i am not sure how to ensure that it is functional.

Help will be appreciated

We usually use 20% glycerol in LB media, and it works 100% of the time. 15% may be fine, but you're pushing it a little in a -80C freezer.

glycerol stock ranges from 10% to 25% commonly. for normal cloning work, 15% glycerol is enough, but if it's a expression plasmid, then its best recommended to store them in 10% to maintain stability.
