Default Glucose isomerase assay



Hi
Pls suggest me the assay methods for Glucose/xylose isomerase assay method. We dont have HPLC facility, so pls suggest me spectrophotometric method.

You could use the enzyme to convert D-glucose into D-fructose. Then after boiling to inactivate the enzyme after a set time, say 5 min, use a D-glucose/D-fructose assay kit to measure the amount of each sugar individually.

Kits to do this are available from various manufacturers.

Sigma publish most enzyme assays on their website (see here for an example for a related enzyme: [Only registered users see links. ]), or alternatively if you can't find an assay there, you could try the specialist recombinant enzyme manufacturer Prozomix ([Only registered users see links. ]). This company is always very helpful when it comes to email queries regarding uncommon assays and assay development.
