Default Plasmid Isolation

Hi
i need information regarding the plasmid DNA isolation that are more then 100kb in size. what is the principle and procedure of isolation.
if some review or article is there then it will be more informative for me.

Hey Aftab,

For a huge plasmid like that many have used cesium chloride gradients. However these arent as clean as other methods and it takes more time than using a commercial kit. However, cesium chloride gradients may be your best change for isolating really clean, large plasmid DNA.
