Default ELISA Practical

Hi there,
Can anyone please tell me the reason for using MArvel milk powder with PBS-Tween to wash the plate. And why would you use two different concentrations e.g 3% MArvel in PBS-Tween and then 0.1% Marvel in PBS-Tween?

Thanks in advance for any help
