| DNA fingerprinting = ribotyping however probes are used that are specific for conserved domains of ribosomal RNA coding sequences these are used to detect the band patterns this will allow the detection of different species RNA |

| Dear Aftab Genes for the Ribosomal RNA are thought to be relatively conserved. If we sequence the 16S ribosomal RNA gene of our unknown isolate and compare it with database of already sequenced ribosomal RNA genes from different microbes, we can identify our unknown isolate on the basis of ribosomal RNA gene homolgy as such databases are very large. outline of te protocol is as follow: Isolate DNA from your unknwn strain. Design primers in the region of 16S RNA gene or use already published set of primers. Get them synthesized fro some commercial source. Amplify 16S RNA gene using these primers and isolated DNA. You may need to gel purify your amplified fragment. sequence the amplified fragment. Search for similar sequences using some search tool (e.g; BLAST) on the net. Try to read some good protocl book for the deatil. Good luck |
