Default Immunostaining of live protoplasts

Hi Arabineters.

I want to find out whether a protein is surface exposed(plasma membrane
bound) or intracellularly expressed in BY2 protoplasts. If I go for
immunostaining, can anyone please suggest any fixing protocol that don't
permeabilize protoplasts?Alternatively if I want to go for staining of
live protoplasts, can anyone suggest any useful tips.

Regards & Thanks

Anirban
