| Hi Everyone, Does anybody have a method for screening SAIL T-DNA lines? I'm an experienced hand at screening other T-DNA lines (SALK, GABI, FLAG), but I'm having real problems with all the SAIL lines i've tried. I'm only able to amplify the wild type size PCR product, even though some plants in my screen are giving an observable phenotype! There's a chance that i've been sent the wrong lines from the stock centre, meaning i'm screening the wrong gene. But i've screened 5 lines and they're all wild type. The chances of the stock centre getting wrong 5 times is pretty slim. Does anybody have any ideas? |
