Default Cleaved caspase-3 positive control in vivo



I am doing Western blot analysis of cleaved caspase-3 in the brain of adult mice. My first western didn't show a signal in any of the treatment groups. Could someone give me an idea of what to use as a positive control to use that I can treat my mice with and see a signal in the brain of mice. I was thinking of using methanesulfonate (MMS) but I am not sure how long I have to wait for the apoptosis to begin.

hi toronto student. i've used HeLa or Jurkat cell lines treated with Staurosporine as positive control groups even though my samples were mouse tissue (can't inject into animals staurosporine). worked fine for me in immunofluorescence microscopy. i'll start western blots soon but i'm not too sure whether I have to use protease or protease + phosphatase inhibitors? do you know or have any idea what the pro/ con are?
