Default comet assay from frozen tissue?



I need to prepare comet assay from frozen cardiac muscle tissue. Is it possible to get some cells from non-cryopreserved tissue in order to perform comet assay?
I`ve tried simple homogenization and collagenase digestion methods- no positive results i.e. no cells on slides.
Any solution?
Help, please...

I have tried this before too but in the brain. Unfortunately you cannot get cell from frozen tissue. Once it is frozen you will not be able to thaw it and get whole cells out.

You have to put fresh samples in the mincing solution, dissociate them and then you can freeze the sample for later analysis.
