jiji... when u say antibody.. u mean monoclonal or polyclonal ab??
Anyway if your peptide size is huge... then u should not be facing much problems obtaining a good titre for your antibody from the host animal..
Tips:
1. Immunize at reasonable concentration and at suitable intervals (2 weeks or 4 depeding on host animal and type of ab).
2. Design ur titre determination protocol exp: ELISA
3. Store ur serum at -20C after harvest.. do not purify it immediately. Jst aliquot it to smaller fractions for storage.
4. Avoid freeze thaw cycles.

Always ensure you use a peptide that is a minimum of 85% pure! certainly in my experience some of the polyclonal suppliers make them as low as 50 and 60% pure which makes for very bad polyclonals.

Just remember to ask... most good companies will advise you correctly.

You should contact a peptide company to do an antigenicity analysis. Many offer this for free. The key is to visit the peptide companies that do the design, because although visiting an Ab company first is intuitive, the antigen you choose - and the quality is the important part. Global Peptide is a good souce.

antigenicity analysis is a very important step I agree Rdonoho! if your peptide is not antigenic this is a problem for antibody generation...

Hello:
We have an issue with the antibody generated against custom syntesized peptide. We have got good results with immunocytochemistry experiments, where as under SDS-PAGE we haven't get the right molecular size as it's supposed to. Can anyone explain this? I dont know where is the problem lies?
