Hi
I am a newbie and wondered if anyone could answer the following question please:

Why should you expect discreet bands cutting lambda dna with HindIII and a smear when cutting plant dna with the same enzyme?

The lambda dna length is about 48 kb. The one of plants differ enormously (Genlisea margaretae genome ist about 63 million bp and fritillaria assyrica genome is about 130 billion ! base pairs) but normaly in contrast to lambda, plant dna much greater. So if you cut with HindIII in suggestion both dna has got nearly the same distribution density (in average 48 kb of plant dna has got the same count as lambda) of restriction sites included then in case of genlisea for example there would be about 1400 times more bands generated. So the smear results in the high number of bands generated by cutting plant dna because of its length.
