Lightbulb Reusing TAE Buffer for Agarose Gel Electrophoresis?



Hi is it possible to reuse your TAE buffer for Agarose Gel Electrophoresis?

My questions are:

  1. Does the primer / DNA that runs of the gel affect later gels?
  2. Does the buffer salts get lost somehow?
  3. Do the dyes (running and loading) affect later running?
  4. What about Ethidium? How to stain later gels?

Hi

you can use TAE buffer more then one times and it usually dont distrub your results.
for routine you can do run the gel in same buffer many times but if you have critical experiment then you should use fresh buffer each time.

there are many ways to stain the gel, you can put ethidium bromide in get during its solidification, if you want to use your TAE buffer again and again.

The general rule of thumb we use here for adding eth. bromide into the gel is to heat it up, then let it cool off until you can handle it with your bare hands with out burning yourself. I would say it's probably around 45 to 50 C when we do so.

one thing you can do is to add water to the gel tank and add a little virkon for one hour and then place the gel tank under UV for 5 mins before using it, especially when you are doing gel purification for ligation purposes because sometimes, the little DNA in the buffer left in the gel tank can run into the gel and be purified together with your DNA. this is what I do whenever I am doing critical steps.

and, I usually don't use my buffer more than 5 times..
