isolation of plasmid

Hi all,
I ran a gel of isolated plasmids into 8 wells.4 of the wells gave bright glow colour which indicates the abundance of plasmids (am i right?).4 others gave faint uniform bands (looking as though they were digested even though they are not).pls i dont understand.if u want me to send the pictures i would.Help

you can post your gel picture here by attaching the picture (go Advanced - button below) then Attach browse to your picture and click submit.

You may have bad DNA preps for those 4 or DNAse contamination / degradation of your DNA.
Did you isolate them differently or store them differently?

i am sorry for my late reply.i actually did not know how to operate the reply thread until now.i am sending the attachment.pls when the lanes have a convex end instead of a straight one.is it significant.Remember i have neither digested or ligated them.i just put them straight in the Ecoli.Can i detect shapes of the plasmids from these lanes.Thanks
